A positive Phase 3 trial results has been announced for a new oral medication (ritlecitinib), reporting that the drug helped restore skin pigmentation in people with nonsegmental vitiligo, including patients with longstanding or advanced disease. The company plans to submit data to health authorities for review.
The clinical programme met its primary and key secondary endpoints, demonstrating statistically significant improvements in skin repigmentation compared with placebo. One primary measure was the Facial Vitiligo Area Scoring Index (F-VASI), which evaluates facial pigmentation improvements. Patients receiving it achieved significantly greater facial repigmentation than those receiving placebo, with improvements becoming more pronounced over the course of treatment.
Encouraging responses were also observed among patients with more advanced or longstanding disease, an area where treatment options have traditionally been limited. The drug’s safety profile was generally consistent with previous studies of Litfulo for alopecia areata, for which it is already approved in several countries.
Vitiligo is a chronic autoimmune condition in which the immune system attacks melanocytes, the cells responsible for producing melanin. As melanocytes are lost, patches of lighter or white skin develop, often appearing symmetrically on the face, hands, and arms. Although not physically painful, vitiligo can have significant psychological and social impact.
It is a Janus kinase (JAK) inhibitor, working by interfering with immune signalling involved in autoimmune inflammation. Current vitiligo treatments include topical corticosteroids, topical JAK inhibitors such as ruxolitinib, calcineurin inhibitors, and phototherapy.
If approved, it could become the first oral treatment specifically authorised for nonsegmental vitiligo, potentially offering an important option for patients who have not responded adequately to existing therapies.
